How to prepare specimens for regular
transmission electron microscopy
Cut the living worms by a blade in the fixative, one by one. Needs patience.
Do not use dead worms which were killed by the fixative.
And then follow the regular method for electron microscopy.
Adults & Newborn larvae
Follow the regular method for electron microscopy.
The fixative can penetrate in through the cuticle.
How to prepare specimens for immunoelectron microscroscopy
Fix worms following the method described above.
Embedd the worms in LR White following the regulasr method.
Cut the section and do immunostaining.
After the immunostaining, put formvar membrane for mechanical support, because the LR White section is so fragile that the worm cannot tolerate against electron beam.
